
"Sterilization in Place" or "Steam in Place" (SIP) is an indispensable procedure in pharmaceutical production. It involves the sterilization of the respective process equipment directly in the system using condensing steam.
Sterilization after Cleaning in Place
After the process-specific cleaning procedures ("Cleaning in Place" or CIP), pipelines, devices, and other components of pharmaceutical manufacturing must be regularly and reliably sterilized. By killing any microorganisms such as germs, bacteria, and viruses, the desired product quality and thus patient safety are ensured.
Sterilization without Dismantling
In SIP processes, the sterilization of the process equipment takes place directly in the closed system using steam, without prior dismantling of the individual components. This minimizes the risk of recontamination. At the same time, manufacturers must design their production systems to be SIP-compliant, which means: free of dead spaces, with smooth surfaces, gap-free constructions, and without the risk of air pockets. The materials used must withstand high temperatures. A guideline for this task is the internationally recognized standard ASME BPE ("Bioprocessing Equipment"). Hot Steam and Atmospheric Pressure
SIP is based on steam generated from pure water by a generator. The steam temperatures range between 121 °C and 134 °C under saturated steam conditions with an overpressure between 1 and 2 bar (15 and 30 psi). The sterilization time at the coldest point is at least 20 minutes. However, pressure and temperature in SIP can vary depending on the device design, product sensitivity, and regional regulations. The requirements are set by the relevant regulatory bodies and institutions, such as the "Food and Drug Administration" (FDA) in the USA and the "European Medicines Agency" (EMA) in Europe.
Automation and Validation of SIP Processes
Most SIP processes are automated to ensure consistency and reliability. Appropriate instrumentation monitors the critical parameters of temperature, pressure, and sterilization duration. The SIP process also requires comprehensive documentation of the procedures. Validation of these processes is essential to prove that all sterilization requirements are met and that the underlying regulations are followed. As necessary as the validation of the procedures is the regular inspection and maintenance of the SIP systems. This also includes the calibration of all measuring instruments.
